Buprenorphine is a partial agonist at the μ receptor with a highly potent analgesic activity in widespread
clinical use for acute and chronic pain. Analgesic activity of buprenorphine is 30 times potent than
morphine and it's major advantages over other analgesics are low dependence-liability and less adverse
effects. Buprenorphine has good pharmacological characteristics for the long term application to the
moderate to severe pain, i.e. cancer pain.
Conventional dosing regimen (IM, IV and sublingual) requires multiple daily dosing and chronic patients
have discomfort in taking drug over a prolonged period of time. With the transdermal delivery technology,
we could apply one patch every three days. And there is one transdermal delivery product of
buprenorphine with the matrix type. And in that case, the absorption of buprenorphine after the first
patch application is slow, achieving the minimal effective concentration after 12 to 24 hours and the
maximal concentration in the plasma after 57 to 59 hours. Samyang has developed hydrogel system
that shortens the initial lag time to less than 6 hours. As our system contains water in the gel,
it is very friendly system to the skin.
